Description
Public Health England (PHE) is seeking to procure an external service provider that can assess the chromosomal stability of human induced pluripotent stem cells (iPSCs).
The proposed service must:
- Be able to receive, handle and process live cells that are Advisory Committee on Dangerous Pathogens (ACDP) Hazard Group 2.
- Have experience handling and fixing human induced pluripotent stem cells (iPSCs) e.g. identifying colony cultures that are in the log phase of growth.
- Use Colcemid for arresting cells in metaphase so that chromosomes can be separated for cytogenetic studies.
- Fix the cells using potassium chloride (KCl) solution and 3:1 methanol: acetic acid.
- Assess the karyology of the fixed cells by G-banding - at least 20 G-banded metaphase spreads needed for each sample.
- Provide a summary of the representative karyotype, along with additional information if any karyotypic abnormalities are found
